The study proposes a comprehensive PK/PD model for 6-mercaptopurine (6-MP) used in childhood acute lymphoblastic leukemia (ALL), integrating:
• Metabolite dynamics (6-TGN, 6-mMPN, 6-TU)
• Genetic variability (e.g., TPMT)
• Age-specific pharmacokinetic changes
• Lifestyle-related inflammation (nutrition, diet, vitamin D)
• Suppressive effects on the hematopoietic system
🧬 Simulations across different patient profiles demonstrate meaningful leukocyte responses and provide a foundation for personalised dose optimisation in pediatric ALL treatment.
